A leader of Irish Presbyterianism, Cooke was educated at the University of Glasgow. He became pastor of the Duneane Presbyterian Church, County Antrim, in 1808.
After holding other pastorates, he was appointed to that of the Presbyterian congregation in May Street in Belfast.
