Samuel was the son of George Duffield, Jr., and husband of Harriet Hayward.
He graduated from Yale College in 1863.
In 1866 he was licensed, and in 1867 ordained, as a Presbyterian minister. He served at churches in Bergen, New Jersey; Ann Arbor, Michigan; Chicago, Illinois; and Altoona, Pennsylvania. In 1881 he became pastor of the Westminster Church, Bloomfield, New Jersey, where he stayed until his death.
