Amélie was the daughter of Aimé Humbert-Droz and Marie Müller.
She grew up in Neuchâtel, and in her youth, was active with the Salvation Army. She was private secretary of Josephine Butler (1876–85), opposed the official regulation of prostitution, and followed her mother in the movement for moral recovery.
She served as secretary (1888–1915), then vice-president (1915–25) of the International Union of the Friends of the Girl and editor of its journal, Le Journal du Bien Public.
