Son of Matthew and Sarah Anderson Lowrie, and nephew of Senator Walter Lowrie, John was educated at Lafayette College, Easton, Pennsylvania (1840); Princeton Theological Seminary, Princeton, New Jersey (1843); and Miami University, Oxford, Ohio (DD 1858).

Lowrie was licensed to preach by the Newton Presbytery April 27, 1842, and ordained at Blairstown, New Jersey, October 18, 1843. He pastored in Blairstown and Knowlton, New Jersey (1843-45); Wellsville, Ohio (1846-50); Lancaster, Ohio (1850-56); and Fort Wayne, Indiana (1856-67).
