John was the son of Major Norman Shairp of Houstoun.
His education included the University of Glasgow (1836–39); Snell Exhibitor, Balliol College, Oxford (1840); and Newdigate Prize (1842).
For a time he was assistant master at Rugby, then Professor of Latin at St. Andrews (1861); Principal of the United College of St. Salvator and St. Leonard, St. Andrews (1868); and Professor of Poetry, Oxford (1877).
The University of Edinburgh conferred a LL.D. on him in 1884.
