John was the son of Matthew Lowrie and Sarah Anderson, and nephew of Senator Walter Lowrie.
He was educated at Lafayette College, Easton, Pennsylvania (1840); Princeton Theological Seminary, Princeton, New Jersey (1843); and Miami University, Oxford, Ohio (DD 1858).
Lowrie was licensed to preach by the Newton Presbytery April 27, 1842, and ordained at Blairstown, New Jersey, October 18, 1843. He served pastorates in Blairstown and Knowlton, New Jersey (1843–45); Wellsville, Ohio (1846–50); Lancaster, Ohio (1850–56); and Fort Wayne, Indiana (1856–67).
