Henry was the son of John Mills, and husband of Maria Moore Barkins.
He was educated at New Jersey College, Princeton, where he graduated in 1802.
After teaching for some time in Morristown and elsewhere, he was ordained pastor of the Presbyterian Church in Woodbridge, New Jersey, in 1816.
After the opening of New York’s Auburn Theological Seminary in 1821, he was appointed Professor of Biblical Criticism and Oriental Languages there, retiring in 1854.
